neoSwitch™ flex workstation
Built for the neoSwitch Antibody Discovery Platform
The Opentrons Flex™ neoSwitch™ Workstation is built for Neochromosome’s neoSwitch antibody discovery platform to automate multiple aspects of the antibody discovery process. neoSwitch toggles between surface display and secretion of hits, and when combined with the Opentrons Flex neoSwitch workstation, those hits can be automatically purified in 24 or 96 well plates. Rather than spending multiple days to manually purify 24 hits, users can drop neoSwitch supernatant on the Flex and return 2 hours and 15 minutes later to retrieve the purified proteins for downstream analysis. Other automation-friendly steps include protein quantification and genotype identification in each clone via colony PCR and library preparation for sequencing.
High Throughput Characterization Antibody Discovery Flex workstation to unlock workflow automation with neoSwitch specific workflows:
- HTP purification of antibodies from neoSwitch supernatant
- Sanger sequencing and library prep
- BCA, ELISA, and other characterization assays
Automated purification of 96 proteins from neoSwitch in 2 hours on the Opentrons Flex.
Magnetic Ni-NTA Flexification Workflow
Time estimates for full plate (24 or 96 DWP) with 8 channel pipette.
In addition to being more reliable, purification of 24 or 96 proteins on the Opentrons Flex neoSwitch Workstation is fully walkaway.
Purification of anti-CD5 VHHs from the neoVHH library™
Stain-free PAGE gel of 8 anti-CD5 VHHs discovered in from the naive neoVHH library in neoSwitch. Left – supernatant prior to purification. Right – Ni-NTA purified anti-CD5 VHHs.
